氣相色譜法同時測定夜寒蘇藥材揮發(fā)油中α—蒎烯、β—蒎烯及芳樟醇的含量

  摘 要 目的:建立同時測定夜寒蘇藥材揮發(fā)油中α-蒎烯、β-蒎烯及芳樟醇含量的方法。方法:按照2015年版《中國藥典》(四部)水蒸氣蒸餾法進行夜寒蘇揮發(fā)油的提取。采用氣相色譜法測定含量,采用HP-5毛細管色譜柱(30 m×0.032 mm×0.25 μm),氫火焰離子化檢測器(FID),載氣為氮氣,進樣口溫度為200 ℃,檢測器溫度為250 ℃,柱流量為0.8 mL/min,進樣量為1 μL,分流比為40 ∶ 1,程序升溫。結果:α-蒎烯、β-蒎烯及芳樟醇的線性范圍分別為0.090 5~2.413 3 mg/mL(r=0.999 9)、0.098 3~2.620 0 mg/mL(r=0.999 9)、0.169 1~4.510 0 mg/mL(r=0.999 8),精密度、穩(wěn)定性(12 h)、重復性試驗的RSD≤2.0%(n=6),平均加樣回收率分別為99.84%(RSD=0.49%,n=6)、100.24%(RSD=1.38%,n=6)、99.41%(RSD=1.67%,n=6)。23批黔產夜寒蘇樣品揮發(fā)油中α-蒎烯、β-蒎烯及芳樟醇含量分別在0.214 4~1.325 0、0.766 2~3.172 1、0.357 4~1.518 7 mg/g范圍內。結論:本試驗建立的方法快速、簡便、準確,可用于夜寒蘇藥材揮發(fā)油中α-蒎烯、β-蒎烯及芳樟醇含量的測定。

  ABSTRACT OBJECTIVE: To establish a method for simultaneous determination of α-pinene, β-pinene and linalool in volatile oil of Hedychium flavum. METHODS: The volatile oil was extracted from H. flavum according to steam distillation stated in Chinese Pharmacopoeia (2015 edition volume Ⅳ). GC method was adopted to determine the content. The determination was performed on HP-5 capillary chromatographic column(30 m×0.032 mm×0.25 μm)with hydrogen flame ion detector (FID), nitrogen gas as carrier gas, with injector temperature of 200 ℃, and detector temperature of 250 ℃, column flow rate of 0.8 mL/min, samples size of 1 μL and split ratio of 40 ∶ 1 by programmed temperature. RESULTS: The linear range of α-pinene, β-pinene and linalool were 0.090 5-2.413 3 mg/mL(r=0.999 9), 0.098 3-2.620 0 mg/mL (r=0.999 9), 0.169 1-4.510 0 mg/mL(r=0.999 8), respectively. RSDs of precision, stability (12 h), reproducibility tests were no more than 2.0% (n=6). The average recoveries were 99.84%(RSD=0.49%, n=6), 100.24%(RSD=1.38%, n=6), 99.41% (RSD=1.67%, n=6), respectively. The contents of α-pinene, β-pinene and linalool ranged 0.214 4-1.325 0, 0.766 2-3.172 1, 0.357 4-1.518 7 mg/g in volatile oil from 23 batches of H. flavum produced in Guizhou province. CONCLUSIONS: The method established in this experiment is rapid, simple and accurate. It can be used for the content determination of α-pinene, β-pinene and linalool in volatile oil from H. flavum.

  夜寒蘇為姜科植物黃姜花(Hedychium flavum Roxb.)的干燥塊根,性溫,味微甜,無毒,具有溫中散寒、解表發(fā)汗、祛風除濕、補虛等功效[1-4]。夜寒蘇是貴州省苗族、布依族、侗族等同胞習用的藥物。在20世紀70年代,《貴州草藥》記載了夜寒蘇在貴州民間常將該藥用于治療寒濕白帶、體虛自汗、風濕筋骨疼痛、消化不良、感冒、胃氣虛弱、頭痛身痛、跌打損傷等病癥[5]。楊秀澤、周漢華等[6-7]雖然已對夜寒蘇進行了性狀及化學成分鑒別,但夜寒蘇化學成分的研究報道目前相對仍然不足,其主要成分是揮發(fā)油。目前夜寒蘇尚未被《中國藥典》《地方標準》收載,沒有明確的質量標準。鑒于此,筆者依據《貴州省中藥材、民族藥材質量標準》(2013年版)中規(guī)范和要求,采用氣相色譜法同時測定夜寒蘇藥材揮發(fā)油中α-蒎烯、β-蒎烯及芳樟醇含量的方法。
